I would say your joruney too tight
try to minimise the place or extend the stay

As what other said

I suggest u stay at least 3 days in Rome

The old city + colosseum take you one whole day

Vatican city + St Peter Church + Museum including queueing another day

You still have other Rome site to see such as Pantheon etc
